If I use the SetSytemSetting() function to set IP settings (i.e. IP Address, Gateway, ...) at runtime, will they take effect immediately, or will a restart be required? If they take effect right away, how can I tell when it is safe or ready to start using the ethernet port for communication again?

That depends on whether you specify SYSACT_DONOW or SYSACT_ONBOOT

In general, I tend to recommend that you reboot afterward as changing the IP address will close any existing network connections, but many objects and code are not written to expect this to happen to them.

If you do elect to change the IP address on-the-fly, you should be able to use the new IP immediately.
